Work Flow:
Bacterial cells are lysed under alkaline conditions. The Endotoxin Removal Buffer is added to the filtered lysate and incubated on ice. The cleared lysate is then loaded onto the anion-exchange tip. After wash, RNA and proteins are removed. Ultrapure endo-free plasmid DNA is eluted in high-salt buffer. The DNA is concentrated and desalted by isopropanol precipitation and resuspended in endo-free TE buffer. DNA yields are up to 500 µg (Maxi-prep), 2.5 mg (Mega-prep), or 10 mg (Giga-prep). The purified DNA is suitable for transfection-grade applications.
